Dionex RF2000 Fluorescence Detector

In addition to the General Device Commands, the RF2000 fluorescence detectors support the commands listed below.

Command/Property

Description

AcqOn/AcqOff

Start/stop data acquisition.

Autozero

Performs automatic null balancing (= the current detector signal is interpreted as 0).

Average

Average all measured values over the Step interval.

Delta

Indicates the signal slope, that is, the difference between the current value and the value one second ago; this is useful for triggers.

EmWavelength

Sets the measuring wavelength. To be able to measure the light emitted by the substance, set the fluorescence detector exactly to the corresponding wavelength.

EventMark

Adds an event mark to the analog output.

ExWavelength

Sets the excitation wavelength. This is the wavelength required to induce a specific substance to fluoresce.

Gain

Determines the factor by which the analog output signal of the spectrometer is increased or decreased. Thus, the Gain parameter corresponds to the Range parameter that is supported for other detector types.

 Note:

For detectors with analog data acquisition, the UCI Universal Chromatography Interface ranges from -10V to +10V and thus covers virtually all detector output signals. It is possible to record signals directly, without considering the Gain parameter. If size adjustment is necessary anyway, adjust the size, using the Factor parameter of the signal configuration. (Select the Signals tab page of the fluorescence detector properties in the Server Configuration program.)

Lamp

Reports the lamp's state: Off, On.

LampAge

Provides information on the lamp's total operating time in [h] (read-only).

LampOn/Off

Turns the lamp on/off.

MaxAutoStep

Sets the maximum step if Step = Auto.

Response

Response is a filter parameter.

A larger time interval improves the Signal-to-Noise Ratio, but lowers the resolution. Use low values for narrow peaks:

Retention

Reports the signal's retention time (read-only).

Sensitivity

Determines the detector sensitivity. The detector sensitivity increases with a higher Sensitivity value. However, increasing the Sensitivity value also increases the noise; that is, the Signal-to-Noise Ratio decreases. Values: LOW, MED, and HIGH. In the HIGH position, the detector sensitivity is approximately 30 times higher than in the LOW position.

The actual sensitivity results from the product of the Sensitivity (LOW = 1, MED = 32, HIGH = 1024) and the Gain (1/4/16).

Signal

Reports the current signal value [mAU] (read-only).

Spectra scan commands and properties

In addition, the RF2000 detector supports the Scan Mode for recording emission and excitation spectra. For more information, refer to Practical Tips for Device Control  Recording Fluorescence Spectra. For a PGM File example, refer to  Determining the Optimum Emission Wavelength (RF2000).

Retrieves a difference spectrum.

SaveBackgroundSpectrum

Saves a background spectrum separately in the detector.

ScanEmission

Performs an emission spectrum measurement. The result spectrum is stored in a spectra library.

ScanEndWavelength

Determines the upper limit of of the wavelength range to be scanned. See also ScanStartWavelength.

ScanExcitation

Performs an excitation spectrum measurement. The result spectrum is stored in a spectra library.

Scanning

Scanning indicates that the detector is currently in the Scan Mode. The Scan Mode functionality determines optimum values for the excitation and emission wavelengths.

ScanSpeed

Sets the speed with which the determined wavelength range is scanned. The following settings are available:

ScanStartWavelength

Determines the lower limit of the wavelength range to be scanned. See also ScanEndWavelength.

ScanWithAnalogOutput

Enable Scan With Analog Output to display the scanned spectrum at the integrator or recorder output. On the instrument's rear panel, these outputs are labeled as follows: INTEG.1V and REC. 10mV or REC. 1mV.
