Sequence Wizard: Methods and Reporting
In this step of the Sequence Wizard (see Samples and Sequences The Sequence Wizard), determine the Chromatographic Methods for all samples.
Use Application Template: |
Select this check box to use an application template for data acquisition and analysis. See About Application Templates for details about the templates. When you select this check box (and a suppressor is installed in the system) the Suppressor Type list appears. Select the type of suppressor installed. Note that this selection has no effect on the actual suppressor configuration; the purpose is simply to display a list of application templates for a particular suppressor. After selecting the suppressor (if any), select the application from the list. The devices configured in the system (pump, detector, eluent generator (if any), and suppressor (if any) determine the applications shown in the list. If you do not select this check box, you can specify individual programs and quantification methods (see Program and Quantification Method below). |

Program: |
Select the Program to be used for processing standard and unknown samples. Click the Browse button to select a program from a different directory. (The Browse button is not available for Chromeleon Xpress.) When the Sequence is generated, the program selected here is automatically copied to the sequence (specified in Step 5 of the Sequence Wizard). Tip: Because a copy of the program selected in this field is saved in the sequence, you can change the copied program without affecting the original program, and vice versa. |

When this check box is selected, a list of the programs previously created for the system is enabled. Select one or more programs to be copied into the sequence. After you load the sequence into the batch, these programs can be used to run the samples in the sequence. See Chromeleon Xpress: Running Samples |

Quantification Method: |
(Optional) Select the method (Quantification Method) to be used for quantitative and qualitative analysis. Click the Browse button to select a method from any directory. When the sequence is generated, the quantification method selected here is automatically copied to the sequence (specified in Step 5 of the Sequence Wizard). Tip: Because a copy of the quantification method selected in this field is saved in the sequence, you can change the copied quantification method without affecting the original quantification method, and vice versa. |

Preferred Report: |
(Optional) If you enter or select a Report, this report definition will automatically be used when opening a sample of the corresponding sequence. In the Batch Report dialog box, this report will be available as the default setting. To change the Preferred Report, select the corresponding sequence in the Browser and select the Properties command. |

Preferred Channel: |
(Optional) If you select a preferred channel, this Channel is used as the default in the Browser and is opened by double-clicking a sample. In the Print Report dialog box, this channel will be available as the default setting. To change the Preferred Channel, select the corresponding sequence in the Browser and select the Properties command. |
Each application template consists of a program and a quantification method (written by Dionex) with the recommended settings for a particular ion chromatography system and column. To see a complete list of available application templates for all devices, see IC Devices Supported by Application Templates.
When the sequence is generated, Chromeleon creates the appropriate program and quantification method (based on the selected template) and copies them to the sequence (specified in Step 5 of the Sequence Wizard). If a control panel appropriate for the timebase exists, it is also copied to the sequence.
The timebase configuration determines which control panel is selected, not the application template. For example, the same control panel will be copied to the sequence for a timebase with a GP50 and a CD25, regardless of whether you select the AS11 or AS12A Production Test Chromatogram template.
Because copies of the template program and quantification method files are saved in the sequence, you can change the copied files without affecting the originals. It is not advisable to edit the program because it contains the default parameters recommended and tested by Dionex. However, the quantification method is intended only as a starting point for quantification. Because the type of analytes and their retention times vary from system to system, Dionex recommends that you fine-tune the method after running the first injection.
If no autosampler is found in the timebase, Chromeleon assumes that an AS40 Autosampler is connected to the Pump's Relay 1. If, however, manual injection is being used, you must modify the program created by the application template.
