Sample Preparation Options

Use this dialog box to define a group of AS-DV commands to be used for preparing the sample before injection. This dialog box is included in the Program Wizard (see  The Program Wizard) and the Program (PGM) Editor (see  The PGM Editor).

To use this dialog box:

  1. Select the Loading Mode (Loop to load a sample loop or Concentrator to load a concentrator column).

  2. If required for your application, modify the General Parameters. For details, see Sample Preparation Options (AS-DV): General Parameters.

  3. Select the Edit Mode. Refer to the following table to determine the mode to select.

Use Basic mode if you want to …

Use Advanced mode if you want to …

Deliver sample from the vial specified in the sequence.

Override the sequence and deliver sample from a specified vial.

Deliver one of the following sample volumes: volume specified in sequence, full vial, 1/2 of a 5 ml vial, 1/3 of a 5 ml vial.  

Deliver a different volume than is available in Basic mode. The volume can be specified in 100 ml increments.

Deliver rinse from the rinse vial range specified in the Server Configuration program. Refer to Options (AS-DV Autosampler Dialog Box).

Deliver rinse from any vial.

Perform a bleed with the injection valve or the auxiliary valve.

Use the optional AS-DV valve for a function other than injection or bleed.

 Note:

The injection valve is either the optional AS-DV valve configured as an injection valve, or the injection valve in the chromatography system module. The auxiliary valve is the optional AS-DV valve configured as an auxiliary valve.

  1. For Basic Edit Mode, select the Volume of sample to deliver from the vial, select a Bleed option, and enable or disable a Rinse. For details, see Sample Preparation Options (AS-DV): Basic Mode.

  2. For Advanced Edit Mode, add, delete, or modify commands and parameters in the Command/Parameters table.  For details, see Sample Preparation Options (AS-DV): Advanced Mode.

  3. (Optional) If you selected From Sequence for the sample delivery volume, use the Total Volume Delivered Estimator to calculate the volume of sample that the AS-DV will deliver to the loop or concentrator column.

Total Volume Delivered Estimator

When From Sequence is selected for the sample delivery volume, Chromeleon uses the formula shown in the Total Volume Delivered Estimator to determine how much sample to take from the vial. To calculate the total volume, enter the volume of the loop installed on the injection valve.
