Program Wizard / Program Editor:
Sample Preparation Options: Basic Edit Mode
Details about the Basic Edit Mode available in the AS-DV Sample Preparation Options dialog box are provided below. For an overview of the dialog box, see Sample Preparation Options (AS-DV).
Basic Edit Mode Overview
When you select Basic Edit Mode, a default list of commands is displayed in the Command/Parameters table. The AS-DV executes the commands in the order in which they appear in the table:
LoadPosition: Switches the injection valve to the load position.
DeliverSample: Delivers the injection volume to the loop. By default, the AS-DV uses the injection volume and vial position specified in the sequence.
EndSamplePrep: Identifies the end of the group of sample preparation commands. This command is used when sample overlap operation is enabled in the Server Configuration program. To enable sample overlap, see Options (AS-DV Autosampler Dialog Box).
Inject: Injects the sample.
You can modify the list of commands by selecting among several options (Volume, Bleed, Rinse). When you select an option, Chromeleon adds the appropriate commands and parameters to the Command/Parameters table. For details about each option, refer to Basic Edit Mode Options below.
Basic Edit Mode Options
Option |
Description |
Volume |
Specify the volume of sample to deliver from the vial.
|
Bleed |
Bleed is used to deliver excess sample (and any trapped air) from the vial to waste before delivering the injection volume to the loop or concentrator column. This ensures that only the specified injection volume of sample is delivered to the loop or concentrator column. The bleed occurs only on the first injection from the vial. To disable the bleed, select None. To enable the bleed, select the valve that will perform the bleed and the valve position that will direct the bleed to waste.
Tip: If the valve is plumbed according to the standard plumbing described in the AS-DV operator's manual, select InjectPosition (if you are using an injection valve) or AuxValve.A (if you are using an auxiliary valve). The selected valve command and a DeliverSample Volume = Bleed command are added to the Command/Parameters table. |
Rinse |
To run a rinse cycle before the injection, select the Rinse check box option. |
Rinse Volume |
Enter the desired Rinse Volume. The volume is taken from the vials in the Rinse Vial Range, specified in the Server Configuration program. To define a rinse vial range, see Options (AS-DV Autosampler Dialog Box). An InjectPosition command and a DeliverRinse command are added to the beginning of the Command/Parameters table. |
