Program Wizard/PGM Editor:
Collector - Tube Change Options

In this step of the Program Wizard, define the tube change options for fraction collection with a WPS-3000FC autosampler and WPS-3000PL autosampler with Micro Fraction Collection Option.

Tray type

Indicates the currently installed tray type for the segment of the selected first fraction position (read-only).

Vial type

Select the type of vials used for fraction collection.

Customize fraction height and/or needle extension values

Select this check box if you want to customize the fraction height (for all vial and well types) or the needle extension (for uncapped vials and wells). Type new needle extension and fraction height values in the boxes. For details, also refer to Commands and Tips for Fraction Collection).

Note that as a rule, the default values will be suitable for most applications.

Needle extension

(Uncapped vials only) The needle extension allows you to specify how far the needle protrudes out of the puncturer during fractionation. This is required to prevent eluent or sample from accumulating in the puncturer.

Example: If you set the value to 10000,  the needle will remain 10 mm outside the puncturer.

 Caution:

If you set the NeedleExtension above 11000, the fused silica needle will break when changing the fraction position.

Fraction height

The Fraction Height defines the needle height during fractionation, that is, the distance between the fractionation needle and the bottom of the tube.  Reduce the fraction height to move the needle to the bottom of the vial or well, for example,

  • for continuos fractionation into 384 well plates, preventing air bubbles from being trapped at the bottom of the well.

  • when fractionating at low flow rates, preventing the droplets from sticking on the needle.

Fraction valve operations

(WPS-3000FC only)

 Caution:

To avoid accidental contamination of empty tubes, always accept the Drain/collect on tube change option when using the RowByRow or ColumnByColumn meandering schemes.

Drain/collect on tube change

Click to switch the fractionation valve to Drain between each tube change to avoid spillages (default).

Drain/collect on tray change only

Click to switch the fractionation valve to Drain only when moving from one tray to the next. Use for (deep) well plates where valve switching between the valve during fractionation is not required. The flow rate should not exceed 0.3 mL/min when 384 (deep) well plates are used.

Do not drain/collect

Click to deactivate switching the fraction valve to Drain after each tube or well. Only use with very low flow rates, where spillages are not expected.

This page is provided in the Program Wizard (see Control  The Program Wizard) only if the Fraction Collection driver has been installed in the Server Configuration Program and if Fraction Collection Mode is not set to Off on the Fraction Collection - General Options page of the PGM wizard.
